SQLE过表达对宫颈癌Hela细胞增殖和迁移及侵袭能力的影响

目的 探讨角鲨烯环氧化酶(SQLE)在宫颈癌Hela细胞中的表达及对宫颈癌细胞增殖、迁移和侵袭的影响及潜在机制.方法 选取Hela细胞和H8细胞进行研究,通过慢病毒构建SQLE过表达稳转细胞株,实验分为H8组、Hela组和Hela-SQLE组;采用细胞计数试剂盒(CCK8)检测各组细胞增殖能力;实时荧光定量聚合酶链式反应(qRT-PCR)检测SQLE、Vimentin、E-cadherin、α-SMA mRNA表达;Transwell实验检测细胞迁移和侵袭力;蛋白质印迹实验检测SQLE、Vimentin、E-cadherin、α-SMA 蛋白表达.结果 CCK8 实验结果显示,24、48 和72 h时 H8 组、Hela 组和 Hela-SQLE 组增殖率差异均有统计学意义,F值分别为80.450、140.900和430.100,P值分别为0.003、0.006和<0.001.Transwell迁移实验结果显示,H8组、Hela组和Hela-SQLE组迁移细胞数分别为(28.43±5.22)、(100.43±5.56)和(139.29± 7.61)个,H=17.840,P<0.001.Transwell侵袭实验结果显示,H8组、Hela组和Hela-SQLE组侵袭细胞数分别为(30.43±1.99)、(92.43±9.29)和(129.29±11.80)个,H=17.850,P<0.001.qRT-PCR 结果显示 H8 组、Hela 组和Hela-SQLE 组 SQLE mRNA 相对表达水平分别为 1.00±0.14、6.32±0.83 和 153.17±10.27,H=7.200,P=0.004;Viementin mRNA 相对表达水平分别为 1.00±0.10、6.94±0.53 和 12.32±0.84,H=7.261,P=0.004;α-SMA mRNA相对表达水平分别为 1.00±0.18、3.39±0.40 和 5.77±0.34,H=7.261,P=0.004;E-cadherin mRNA 相对表达水平分别为 1.00±0.07、0.07±0.01 和 0.03±0.00,H=7.261,P=0.004.蛋白质印迹实验结果显示 H8 组、Hela 组、Hela-SQLE 组 SQLE 蛋白相对表达水平分别为 0.15±0.03、0.45±0.02 和 0.64±0.05,H=4.571,P<0.001;Viementin 蛋白相对表达水平分别为0.08±0.03、0.19±0.02和0.35±0.03,F=54.220,P=0.006;α-SMA蛋白相对表达水平分别为 0.06±0.01、0.17±0.00 和 0.45±0.01,F=948.000,P=0.005;E-cadherin 蛋白相对表达水平分别为 0.41±0.01、0.20±0.04和0.08±0.00,F=115.800,P=0.033.结论 宫颈癌细胞中SQLE表达升高,SQLE过表达促进了宫颈癌细胞的增殖、侵袭和迁移能力,可能是潜在的宫颈癌促癌因子.
Effect of overexpression of SQLE on the proliferation,migration,and invasion ability of cervical cancer Hela cells
Objective To investigate the expression of squalene epoxidase(SQLE)in cervical cancer Hela cells and its effect on the proliferation,migration and invasion of cervical cancer cells and its potential mechanism.Methods Hela and H8 cells were sellected for this research,SQLE overexpression stably transfected cell lines were constructed by lentivir-us,the experiment was divided into H8 group,Hela group and Hela-SQLE group.Cell proliferation was detected by CCK8 assay.The mRNA expressions of SQLE,Vimentin,E-cadherin,and α-SMA were detected by qRT-PCR.Cell mi-gration and invasion were detected by Transwelll.Western blotting was used to detect the protein expression of SQLE,Vimentin,E-cadherin,and α-SMA.Results The results of CCK8 experiment showed that the proliferation rate of H8 group,Hela group and Hela-SQLE group was statistically significant at 24,48 and 72 h,with F values of 80.450,140.900 and 430.100,and P values of 0.003,0.006 and<0.001,respectively.The results of Transwell migration ex-periment showed that the number of H8 cells,Hela cells and SQLE overexpressed Hela cells migrated was(28.43± 5.22),(100.43±5.56)and(139.29±7.61),H=17.840,P<0.0001.Transwell invasion assay showed that the number of H8 cells,Hela cells and SQLE overexpressed Hela cells invaded was(30.43±1.99),(92.43±9.29)and(129.29±11.80),H=17.850,P<0.0001.qRT-PCR results showed that the relative expression levels of SQLE mRNA in H8 cells,Hela cells and SQLE overexpressed Hela cells were 1.00±0.14,6.32±0.83 and 153.17±10.27,H=7.200,P=0.004;The relative expression levels of Viementin mRNA were 1.00±0.10、6.94±0.53 and 12.32±0.84,H=7.261,P=0.004;The relative expression levels of α-SMA mRNA were 1.00±0.18、3.39±0.40 and 5.77±0.34,H=7.261,P=0.004;The relative expression levels of E-cadherin mRNA were 1.00±0.07,0.07±0.01 and 0.03±0.00,H=7.261,P=0.004.Western blot results showed that the relative expression levels of SQLE protein in H8 cells,Hela cells and SQLE overexpressed Hela cells were 0.15±0.03,0.45±0.02 and 0.64±0.05,H=4.571,P=0.0002;The relative expression levels of Vie-mentin protein were 0.08±0.03,0.19±0.02 and 0.35±0.03,F=54.220,P=0.006;The relative expression levels of α-SMA protein were 0.06±0.01,0.17±0.00 and 0.45±0.01,F=948.000,P=0.005;The relative expression levels of E-cadherin protein were 0.41±0.01,0.20±0.04 and 0.08±0.00,F=115.800,P=0.033.Conclusions The expression of SQLE in cervical cancer cells is increased.SQLE overexpression promotes the proliferation,invasion and migration of cervical cancer cells.SQLE may be a potential cervical cancer promoting factor.
