Objective To investigate the mechanism of action of the mechanistic target of rapamycin(mTOR)in the devel-opment,progression and treatment of hepatocellular carcinoma.Methods The Pubmed database was used to search the relevant literature from 2001 to 2023 using"mTOR,liver cancer,inhibitor"as keywords.Inclusion criteria:(1)defini-tion,typing and role of mTOR;(2)animal and clinical studies on mTOR in liver cancer;(3)research progress on mTOR inhibitor.Exclusion criteria:(1)conference abstracts;(2)low-quality literature.Seventy relevant literature were finally included.Results The mechanism of mTOR affecting the progression of hepatocellular carcinoma mainly includes promo-ting the proliferation and metastasis of hepatocellular carcinoma cells,inhibiting apoptosis and autophagy of hepatocellular carcinoma cells,destroying the microenvironment of tumors,and generating drug resistance.mTOR inhibitor treatment of hepatocellular carcinoma is a double-edged sword,which can prolong the patients'survival period,reduce the recur-rence of hepatocellular carcinoma,and resist the resistance of drug,but it can also damage the liver function,and even not be beneficial.Conclusions The mTOR signaling pathway is clinically important for the occurrence,progression and treat-ment of hepatocellular carcinoma.mTOR inhibitors,although having significant advantages in the treatment of hepatocel-lular carcinoma,also have shortcomings.
