目的 总结肿瘤相关巨噬细胞(TAMs)在结直肠癌患者免疫检查点抑制剂(ICIs)治疗中所涉及的相关机制,探讨其在逆转ICIs治疗耐受中的应用前景,以期为相关研究提供参考.方法 以"肿瘤相关巨噬细胞、结直肠癌、免疫检查点抑制剂 耐药"为中 文检索 词,以"tumor-associated macrophages、colorectal cancer、immunotherapy、immune checkpoint inhibitor、drug resistance"为英文检索词,检索PubMed、中国知识基础设施工程(CNKI)和万方数据库2000-01-01-2023-04-01发表的相关中英文论著.纳入标准:(1)TAMs与ICIs治疗;(2)结直肠癌与ICIs耐药;(3)靶向TAMs在结直肠癌中的临床应用.排除标准:会议摘要、中文学位论文等非正式发表文献.最终纳入符合条件文献37篇.结果 ICIs作为一类广泛应用于临床肿瘤免疫治疗的药物,可作用于细胞上起抑制作用的调节分子——免疫检查点,通过激活免疫系统中多种免疫细胞,以免疫应答的形式杀灭肿瘤.不同类型的结直肠癌患者对ICIs治疗的敏感性差异很大.结直肠癌肿瘤微环境中TAMs的表型及功能与ICIs的疗效密切相关,ICIs能调控TAMs的表型功能,TAMs也能影响结直肠癌对ICIs治疗的耐受.结论 TAMs在ICIs耐药中起重要作用,充分利用此靶点作为治疗策略,有望改善结直肠癌患者的免疫治疗效果和预后.
Research progress of tumor-associated macrophages in immune checkpoint inhibitor resistance in colorectal cancer
Objective To summarize the relevant mechanisms of tumor-associated macrophages(TAMs)in the treatment of immune checkpoint inhibitors(ICIs)in patients with colorectal cancer(CRC),and explore the application prospect of TAMs in reversing ICIs treatment tolerance,in order to provide reference for related studies.Methods The relevant lit-erature were searched from PubMed,China National Knowledge Infrastructure(CNKI)and Wanfang Database from Janu-ary 1,2000 to April 1,2022,using"tumor-associated macrophages,colorectal cancer,immunotherapy,immune check-point inhibitor,drug resistance"as the keywords.Inclusion criteria:(1)TAMs and ICIs treatment;(2)Colorectal canc-er and ICIs resistance;(3)The clinical application of targeted TAMs in Colorectal cancer.Exclusion criteria:conference abstract,Chinese dissertation and other informal published documents.A total of 37 eligible articles were included.Re-sults ICIs,as widely used drugs in clinical tumor immunotherapy,can act on regulatory molecules with inhibitory effects on cells-immune checkpoints-the tumor is killed in the form of immune response,by activating a variety of immune cells in the immune system.The sensitivity of different types of colorectal cancer patients to ICIs treatment varies greatly.The phenotype and function of TAMs in colorectal cancer microenvironment are closely related to the therapeutic effect of im-mune checkpoint inhibitor ICIs.ICIs can regulate the phenotype and function of TAMs,and TAMs can also affect the tol-erance of colorectal cancer to ICIs treatment.Conclusions Tumor-associated macrophages play an important role in im-mune checkpoint resistance.Making full use of this target as a therapeutic strategy is expected to improve the immuno-therapeutic efficacy and prognosis of patients with colorectal cancer.
