LSAMP-AS1/LSAMP通过PI3K/Akt/MDM2/p53信号通路调控胃癌作用研究

目的 探索长链非编码RNA边缘系统相关膜蛋白反义RNA(LSAMP-AS1)和边缘系统相关膜蛋白(LSAMP)基因在胃癌组织中的表达及临床意义.方法 收集川北医学院附属医院胃肠外科2016-01-04-2018-06-20手术治疗的60例胃癌患者癌组织和癌旁组织标本.采用实时荧光定量聚合酶链反应(qRT-PCR)法检测胃癌组织LSAMP-AS1、LSAMP、磷脂酰肌醇3-激酶(PI3K)、鼠双微基因2(MDM2)和p53的表达水平并分析其关系,采用蛋白质印迹法检测LSAMP的表达.胃癌组织和癌旁组织之间LSAMP、LSAMP-AS1表达量符合正态分布,选用配对t检验;不符合正态分布的信号通路分子的表达比较选择Wilcoxon符号秩检验.结果 (1)在胃癌和癌旁组织中,LSAMP-AS 1的表达量分别为1.18±1.39和2.21±2.60,差异有统计学意义,配对t=3.169,P=0.002;LSAMP的表达量分别为1.90±2.78和2.76±3.55,差异有统计学意义,配对t=2.591,P=0.012.(2)LSAMP-AS1表达与LSAMP呈正相关,r=0.740,P<0.001.(3)LSAMP-AS1低表达癌组织中,PI3K表达高于癌旁组织(Z=-1.586,P=0.063),MDM2低于癌旁组织(Z=-2.749,P=0.007),p53低于癌旁组织,Z=-2.372,P=0.018.(4)LSAMP低表达癌组织中,PI3K表达高于癌旁组织(Z=-4.240,P<0.001)、MDM2 低于癌旁组织(Z=-2.749,P=0.00 6),p53 低于癌旁组织,Z=-2.482,P=0.013.结论 (1)LSAMP-AS1和LSAMP可能作为抑癌基因参与胃癌的发生发展;(2)低表达LSAMP-AS 1和LSAMP可作为不良预后指标,为胃癌诊断、预后判断提供重要参考依据;(3)LSAMP-AS1和LSAMP可能通过PI3K/Akt/MDM2/p53信号通路调控胃癌的发生发展.
Regulatory role of LSAMP-AS1/LSAMP in gastric cancer through the PI3K/Akt/MDM2/p53 signaling pathway
Objective To explore the expression and clinical significance of long non-coding RNA limbic system membrane protein antisense RNA(LSAMP-AS1)and limbic system membrane protein(LSAMP)gene in gastric cancer.Methods Cancerous and adjacent non-cancerous tissue specimens from 60 patients with gastric cancer who underwent surgical treat-ment at Department of Gastrointestinal Surgery,Affiliated Hospital of North Sichuan Medical College were collected from January 4,2016 to June 20,2018.Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)was used to dectect the expression levels of LSAMP-AS1,LSAMP,phosphatidylinositide 3-kinase(PI3K),murine double mimute 2(MDM2),and p53 in gastric cancer tissue and their relationships.Western blot was used to detect the expression of LSAMP.The expression levels of LSAMP and LSAMP-AS1 between gastric cancer tissues and adjacent non-cancerous tissues were normally distributed,and paired t-test was used.Wilcoxon signed-rank test was used to compare the expres-sion levels of signal pathway molecules that did not conform to normal distribution.Results(1)The expression levels of LSAMP-AS 1 in gastric cancer and adjacent non-cancerous tissues were 1.18±1.39 and 2.21±2.60,respectively,with statistical significance.The paired t=3.169,P=0.002;The expression levels of LSAMP were 1.90±2.78 and 2.76±3.55,respectively,the difference was statistically significant,and the paired t=2.591,P=0.012.(2)The expression level of LSAMP-AS1 was positively correlated with LSAMP,r=0.740,P<0.001.(3)In low-expression LSAMP-AS1 cancer tissues,the expression of P13K was higher than that of adjacent non-cancerous tissues(Z=-1.586,P=0.063),MDM2 was lower than that of adjacent non-cancerous tissues(Z=-2.749,P=0.007),and p53 was lower than that of adjacent non-cancerous tissues(Z=-2.372,P=0.018).(4)In low-expression LSAMP cancer tissues,the expression of PI3K was higher than that of adjacent non-cancerous tissues(Z=-4.240,P<0.001),MDM2 was lower than that of adjacent non-cancerous tissues(Z=-2.749,P=0.006),and p53 was lower than that of adjacent non-cancerous tissues(Z=-2.482,P=0.013).Conclusions Based on these findings,it can be concluded that both LSAMP-AS1 and LSAMP may function as tumor suppressor genes involved in the development and progression of gastric cancer.Low ex-pression levels of these genes could serve as poor prognostic indicators for gastric cancer diagnosis and prognosis.LSAMP-AS 1 and LSAMP may regulate the occurrence and development of gastric cancer through the PI3K/Akt/MDM2/p53 signaling pathway.
