Regulatory role of LSAMP-AS1/LSAMP in gastric cancer through the PI3K/Akt/MDM2/p53 signaling pathway
Objective To explore the expression and clinical significance of long non-coding RNA limbic system membrane protein antisense RNA(LSAMP-AS1)and limbic system membrane protein(LSAMP)gene in gastric cancer.Methods Cancerous and adjacent non-cancerous tissue specimens from 60 patients with gastric cancer who underwent surgical treat-ment at Department of Gastrointestinal Surgery,Affiliated Hospital of North Sichuan Medical College were collected from January 4,2016 to June 20,2018.Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)was used to dectect the expression levels of LSAMP-AS1,LSAMP,phosphatidylinositide 3-kinase(PI3K),murine double mimute 2(MDM2),and p53 in gastric cancer tissue and their relationships.Western blot was used to detect the expression of LSAMP.The expression levels of LSAMP and LSAMP-AS1 between gastric cancer tissues and adjacent non-cancerous tissues were normally distributed,and paired t-test was used.Wilcoxon signed-rank test was used to compare the expres-sion levels of signal pathway molecules that did not conform to normal distribution.Results(1)The expression levels of LSAMP-AS 1 in gastric cancer and adjacent non-cancerous tissues were 1.18±1.39 and 2.21±2.60,respectively,with statistical significance.The paired t=3.169,P=0.002;The expression levels of LSAMP were 1.90±2.78 and 2.76±3.55,respectively,the difference was statistically significant,and the paired t=2.591,P=0.012.(2)The expression level of LSAMP-AS1 was positively correlated with LSAMP,r=0.740,P<0.001.(3)In low-expression LSAMP-AS1 cancer tissues,the expression of P13K was higher than that of adjacent non-cancerous tissues(Z=-1.586,P=0.063),MDM2 was lower than that of adjacent non-cancerous tissues(Z=-2.749,P=0.007),and p53 was lower than that of adjacent non-cancerous tissues(Z=-2.372,P=0.018).(4)In low-expression LSAMP cancer tissues,the expression of PI3K was higher than that of adjacent non-cancerous tissues(Z=-4.240,P<0.001),MDM2 was lower than that of adjacent non-cancerous tissues(Z=-2.749,P=0.006),and p53 was lower than that of adjacent non-cancerous tissues(Z=-2.482,P=0.013).Conclusions Based on these findings,it can be concluded that both LSAMP-AS1 and LSAMP may function as tumor suppressor genes involved in the development and progression of gastric cancer.Low ex-pression levels of these genes could serve as poor prognostic indicators for gastric cancer diagnosis and prognosis.LSAMP-AS 1 and LSAMP may regulate the occurrence and development of gastric cancer through the PI3K/Akt/MDM2/p53 signaling pathway.
