细胞自噬治疗肿瘤的研究进展

目的 总结细胞自噬治疗肿瘤的研究进展,以期提高对细胞自噬的认识与理解,促进自噬在肿瘤临床和基础领域的应用与发展.方法 使用中国知网数据库、中华医学期刊全文数据库及PubMed检索系统,以"细胞自噬""自噬信号传导""自噬与肿瘤""靶向自噬"为中文关键词,"Autophagy""Autophagy signal transduction""Autophagy and Cancer""Targeted Autophagy"为英文关键词,检索2013-01-01-2024-01-31相关文献.纳入标准:(1)自噬的过程和调节机制;(2)自噬与肿瘤的关系;(3)调节自噬与肿瘤治疗.排除标准:(1)数据不完整的文献;(2)文献研究方法中存在统计学问题的文献以及内容相似或重复的文献.最终纳入文献73篇,其中中文文献4篇,英文文献69篇.结果 自噬与肿瘤的发生、发展和药物抵抗有关.自噬能通过维持细胞内稳态、DNA和基因组的稳定、增强肿瘤免疫治疗抑制肿瘤发生;也能通过保障肿瘤细胞代谢,维持癌症干细胞稳定促进肿瘤发展;在某些情况下自噬过度激活也可引起自噬性死亡.通过药物促进自噬或者抑制自噬可以对肿瘤细胞起到杀伤作用.结论 自噬过程受一系列复杂信号分子的调控,调控机制与肿瘤的病理生理过程存在密切联系.自噬与肿瘤的发病、治疗和预后密切相关,可以作为肿瘤治疗的潜在干预靶点,调节自噬能为肿瘤治疗提供新的策略.
Research progress of autophagy in the treatment of tumor
Objective To review the research progress of cell autophagy in cancer therapy,so as to improve the understand-ing of cell autophagy and promote the application and development of autophagy in the clinical and basic fields of cancer.Methods Searched the CNKI databases,Chinese medical journal full-text database and PubMed databases using the key-words"autophagy""autophagy signal transduction""autophagy and cancer"and"targeted autophagy".The relevant lit-erature was searched from January 1,2013,to January 31,2024.Inclusion criteria:(1)the process and regulatory mech-anism of autophagy;(2)the relationship between autophagy and tumor;(3)regulation of autophagy and tumor therapy.Exclusion criteria:(1)literature with incomplete data;(2)literature with statistical problems in literature research meth-ods and literature with similar or repeated content.Finally,73 articles were included,including 4 Chinese articles and 69 English articles.Results Autophagy is related to the occurrence,development,and drug resistance of tumors.Autoph-agy can suppress tumor development by maintaining cellular homeostasis,DNA and genome stability,and enhancing tumor immunotherapy.It can also promote tumor development by ensuring the metabolism of tumor cells and maintaining the stability of cancer stem cells.In some cases,excessive activation of autophagy can also cause autophagic death.Pro-moting or inhibiting autophagy through drugs can have a killing effect on tumor cells.Conclusions The autophagy process is regulated by a series of complex signaling molecules,and the regulatory mechanism is closely related to the pathological and physiological processes of tumors.Autophagy is closely related to the onset,treatment,and prognosis of tumors,and can serve as a potential intervention target for tumor treatment.Regulating autophagy can provide new strategies for tumor treatment.
