Chemical composition analysis of Hemerocallis hybrida Bergmans based on UHPLC-Q-TOF-MS/MS technology
Objective By adopting ultra high performance liquid chromatography cascade Quadrupole time-of-flight mass spectrometry to analyze the chemical composition of Hemerocallis hybrida Bergmans. Methods The gradient elution was carried out on A Waters HSS T3 column with 0.1%formic acid acetonitrile(A)-0.1%formic acid water(B)as the mobile phase in negative ion mode. The flow rate was 0.4 ml/min,the column temperature was 35,and the injection volume was 4 pl.The chemical composition database of Hemerocallis was established, and the compounds were identified by accurate m/z values, characteristic secondary data,a variety of databases and cleavage pathways in the literature. Results A total of 59 compounds were identified, including 25 flavonoids,22 phenylpropanoids,4 fatty acids,2 alkaloids and 6 other compounds.Among them,12 compounds were first reported in Hemerocallis. Conclusions This method is the first time to systematically, comprehensively and quickly screen and identify the compounds in Hemerocallis hybrida Bergmans, which provides a basis for the development and in-depth study of Hemerocallis hybrida Bergmans.
