对采自贵州省晴隆县的一株野生羊肚菌QL-1进行鉴定,发现其子实体中等大,菌盖圆锥形,以灰褐色为主,具放射状的条棱形成似蜂窝状凹窝,下部边缘中部与菌柄连接;菌柄较长且粗壮,表面有细颗粒或粉粒密布;孢子椭圆形,纵向单行排列4个.基于ITS序列分析QL-1属黑色羊肚菌支系,是一株适宜本地环境的变种.菌丝纯培养结果显示,最适培养温度为16℃,最适pH环境为6,最佳培养基为小麦培养基;菌丝老化速度测试显示QL-1菌丝平均寿命为30 d.
Isolation,Identification and Mycelial Growth Characteristics of One Wild Morchella Strain
Identificating a wild morchella strain QL-1 from Qinglong County,it is found that its fruiting body is medium-sized with a conical cap and a predominantly grayish blown color.Having radiating ribs forming a honeycomb like recess and the lower edge connected to the stipe,the fungal stalk is long and robust with fine particles or powder particles densely distributed on its surface.The spores are elliptical in shape and arranged vertically in a single row with 4 spores.Based on analyzing,the QL-1 Genus in ITS Sequence,which belongs to the Black Morchella Branch,is a variant suitable for the local environment.The results of pure mycelium culture show that the optimal culture temperature is 21 ℃ and the optimal pH is 6 and the optimal culture medium is wheat.The mycelium aging rate test shows that the average lifespan of QL-1 mycelium is 30 days.
