Methylation Analysis of 5S rRNA Gene in the Hybrid Grouper(Epinephelus fuscoguttatus × Epinephelus polyphekadion)
Ribosomal RNA(rRNA)is an important component of ribosomal which plays an important role in protein synthesis.The hybrid grouper derived from the hybridization of Epinephelus fuscoguttatus × Epinephelus polyphekadion,exhib-its a certain of advantages in growth and meat quality.The DNA methylation levels of the first 1 000 bp of the coding re-gion of 5S rRNA gene,coding region and non-transcribed spacer of 5S rRNA gene were measured and analyzed contras-tively between the hybrid grouper and its parents.Results showed that the overall methylation level of 5S rRNA gene in the hybrid grouper(0.418)was higher than that of E.fuscoguttatus(0.379)and E.polyphekadion(0.037).While there was no significant difference between the hybrid grouper and E.fuscoguttatus,significant difference was detected between the hybrid grouper and E.polyphekadion.Comparative analysis of DNA methylation level at each CpG site in the coding region and non-transcribed spacer revealed that all methylation sites co-existed in both the hybrid grouper and E.fuscoguttatus,but were not found in E.polyphekadion.These results indicate that the methylation pattern of the 5S rRNA gene in the hy-brid grouper was similar to that of E.fuscoguttatus.The observations elucidate the DNA methylation level and pattern of 5S rRNA gene in the hybrid grouper,and provide a theoretical basis for cross-breeding in grouper.
