Isolation and Identification of V.cholerae from the Rotting Skin Disease of Paramisgurnus dabryanus ssp.
In order to study the pathogen of P.dabryanus ssp.,a dominant strain named P1 was obtained by bacterial isolation and purification of its rotting skin and other lesions.The morphological characteristics of the strain were observed by Gram staining,and the sequence of the pathogen was obtained by PCR amplification and 16S rRNA gene sequencing.The phylogenetic tree was constructed by MEGA software.Then the name of the bacterial species was identified through physiological and biochemical analysis.The results showed that Gram staining was negative,curved,and bluntly rounded at both ends;the bacterium was 1-1.5 μm in length,1-1.2 μm in width,with mycelium and flagellum;16S rRNA gene was 1 395 bp;the establishment of the phylogenetic tree revealed that P1 was in the same branch with Vibrio cholerae;the results of physiological and biochemical experi-ments were in agreement with those of Vibrio cholerae,which leads to the identification of the strain as Vibrio cholerae.The results of drug sensitivity test showed that the isolated strain was highly sensitive to nine antibiot-ics,including cefoperazone,canomyces and vancomycin,moderate sensitive to tetracycline,resistant to 18 antibiot-ics,including furazolidone,cotrimoxazole and clindamycin.The results provide reference for the pathogen identifi-cation and drug control of P.dabryanus ssp..
