贵州省白术白绢病菌遗传多样性评价

为研究贵州白绢病菌株间的亲缘关系,对采自贵州省贵阳、遵义等5个市(州)郊不同发病地的20株白术白绢病菌进行随机多态性扩增,根据引物扩增的DNA指纹图谱,运用UPGMA法分析其遗传多样性.116条引物中筛选出12条引物,获得了较为清晰的扩增谱带,共获得DNA指纹图带64条,其中多态性条带44条,多态性检测率为68.75%,菌株间相似性系数为0.56~0.98.以遗传相似系数0.79为阈值,供试菌株被划分为3个遗传聚类组(Ⅰ/Ⅱ/Ⅲ).除部分相同地理来源(毕节)的菌株被划分为同一组外,其它不同菌株间的遗传相似性与地理来源直接相关性并不明显.
Genetic Diversity Analysis of Sclerotium rolfsii Sacc.in Rhizoma of Atractylodes Macrocephala
The genetic diversity of Sclerotium rolfsii Sacc. in Atractylodes macrocephala was studied in order to analyse the relationship between the strains from 5 regions (Guiyang, Zunyi, Qiandongnan, Qiannan and Bijie city)in Guizhou Province. 20 strains collected from different biotopes were amplified by 116 RAPD primers. The genetic relationships were generated using UPGMA method of NTSYS software based on the DNA fingerprint. 64 bands including 44 reproducible polymorphic bands with 68.75% polymorphic band frequency based on 12 RAPD primers were obtained . The similarity coefficient between strains was from 0.56 to 0.98, which were divided into 3 groups marked I, II and M based the threshold of similarity coefficient at 0.79. The results indicated that DNA polymorphism had no significant correlation with geographical origins, excepting a few of strains from Bijie were divided into a same group.
