Construction of Molecular Fingerprinting Map of Leaf Mustard [Brassica juncea (L.) Czern et Coss] in China with SRAP Markers
In the study,86 mustard [Brassica juncea (L.) Czern.et Coss.] germplasm from nine provinces in China were used to construct DNA fingerprint using SRAP markers and the DNA fingerprint analysis software.40 pairs of SRAP primers with high polymorphism were selected from 270 pairs of primers,which were used to amplify the genomic DNA of 86 mustard accessions.632 bands were amplified totally,of which 427 bands were polymorphic,and the polymorphic rate was 67.56%,indicating the abundant genetic diversity of mustard accessions.Based on the DNA bands amplified from 86 mustard germplasm resources,the Excel database was established.Furthermore,DNA fingerprints of 86 mustard accessions were constructed using the software designed by our laboratory.The "ID" of each mustard variety was obtained in our study.The result showed SRAP technique is suitable for DNA fingerprint constructing in mustard germplasm.
