Optimization of Extraction Process and Purification Process of Compound Jianpi Yangwei Formula
On the basis of single factor experiment,orthogonal experimental optimization was used to investigate the effects of extraction temperature,extraction time,and liquid to material ratio on the extraction rate of crude polysaccharides in the compound Jianpi Yangwei fomula,and to investigate the effects of purification process on the retention rate,turbidity,and sensory evaluation of crude polysaccharides.The results showed that the optimal extraction conditions of crude polysaccharides were as follows:water extraction at 95℃,the ratio of liquid to material was 12∶1 mL/g,the extraction time was 2 h,and the extraction was twice.The optimum purification process was:the water extract was concentrated to a relative density of 1.05~1.10(65℃),centrifuged at 8 000 r/min for 20 min,and then filtered with a filter bag with a pore size of 0.45 μm.The retention rate of crude polysaccharide in the filtrate was 90.10%,the turbidity was 31.4 NTU,the liquid was clear and transparent,and the stability of the water extract was good.
