颐阳健酒安全性评价研究及对小鼠免疫功能的影响

研究颐阳健酒的食品安全性及对小鼠免疫功能的影响.通过急性毒性实验、30d喂养毒性实验和遗传毒性实验评价颐阳健酒的安全性,通过测定小鼠淋巴细胞增殖能力、DTH程度、溶血空斑数、血清半数溶血值,单核-巨噬细胞功能及NK细胞活性确定颐阳健酒是否具有影响小鼠免疫力功能.结果表明:急性毒性实验结果显示颐阳健酒的雌、雄小鼠急性经口MTD>22400 mg/kg,属实际无毒级;30 d喂养毒性实验结果显示颐阳健酒各剂量组血液学及血生化各项指标均在正常范围;各脏器病理组织学检查均未见与受试样品有关的病理改变.遗传毒性实验结果显示Ames实验、小鼠精子畸形实验和小鼠骨髓细胞微核实验结果均为阴性,未见致畸变作用.功能学研究结果显示,颐阳健酒以0.5、1.0、3.0 mL/(kg·bw·d)三个剂量经口给予小鼠30d后,小鼠淋巴细胞增殖能力、DTH程度、溶血空斑数、血清半数溶血值结果均为阴性,单核-巨噬细胞功能及NK细胞活性均为阳性.该实验表明颐阳健酒食品安全性良好,具有调节小鼠免疫力功能.
Safety Evaluation and Immunoregulatory Effect in Mice of YiYangJianJiu
To evaluate the safety of YiYangJianJiu and its effect on immune functions of mice.The safety of YiYangJianjiu was evaluated by acute toxicity test,30-day feeding toxicity test and genetic toxicity test.The immunity enhancement function of YiYangJianjiu was determined by measuring lymphocyte proliferative ability,DTH level,hemolytic plaques numbers,half hemolytic value of serum,monocyte-macrophage function and NK cell activity of mice.The results of acute toxicity test showed that the acute oral MTD of female and male mice of YiYangJianJiu was more than 22 400 mg/kg,which belonged to the actual non-toxic grade.The 30-day feeding toxicity test showed that the hematological and biochemical indexes of each dose group of YiYangJianJiu were within the normal range.No pathological changes related to the sample were found in the pathological histological examination of organs.The results of genotoxicity test showed that Ames test,sperm abnormality test and micronucleus test of bone marrow cells in mice were negative,and no teratogenic effect was observed.The results of functional studies showed that after oral administration of YiYangJianJiu at doses of 0.5,1.0 and 3.0 mL/(kg·bw·d)for 30 days,the results of lymphocyte proliferative ability,DTH level,hemolytic plaques numbers and half hemolytic value of serum of mice were negative,while the monocyte-macrophage and NK cell activities were positive.YiYangJianJiu is safe as a food supplement and can boost immunity of mice.
