Rapid Determination of Choline in Food for Special Medical Purpose by Enzyme Colorimetry Using Microwave-assisted Hydrolysis
This paper extracted choline from food for special medical purpose using microwave-assisted hydrolysis,and detected the choline content by enzyme colorimetry.Choline was catalyzed by choline oxidase with subsequent discharge of hydrogen peroxide(H2O2).In the presence of peroxidase,hydrogen peroxide reacted with phenol and 4-aminoantipyrine and colored substances were formed.The external standard method was used for quantitative analysis of choline by spectrophotometry.Standard curve,the hydrolysis-solvent and microwave-assisted hydrolysis heating condition were optimized.The determined optimum method was experimentally verified in the samples of different matrices.Selecting samples with 1.0 mol/L HCl solution for microwave-assisted hydrolysis at 100℃ for 5 min,the analysis results and spiked recovery rates agreed quite well with those obtained by the national standard method for water bath heating hydrolysis.The pretreatment method of microwave-assisted hydrolysis is simple,rapid and highly accurate,and it is applicable for batch detection of choline in food for special medical purpose.
