摘要

目的 观察丹参酮ⅡA对成年大鼠睾丸扭转复位后细胞凋亡及性激素水平的影响,探讨其保护作用及机制.方法 将64只雄性SD大鼠随机分为4组,假手术组、模型组、丹参酮ⅡA单次和连续治疗组,每组16只.单次治疗组在睾丸扭转复位后经腹腔注射丹参酮ⅡA(25 mg/kg),而连续治疗组除上述操作外,还在术后1周内1次/d继续注射同等剂量丹参酮ⅡA维持.各组大鼠在再灌注第4天、第29天被处死,采用HE染色和TUNEL荧光染色观察睾丸组织病理学改变,采用Western Blot方法进行检测Bcl-2和Bax蛋白,采用ELISA法检测性激素FSH、LH和T水平.结果 与模型组比较,经过丹参酮ⅡA治疗的大鼠其睾丸组织损伤减轻,生殖细胞凋亡减少,且连续治疗组比单次治疗组改善更明显(P<0.01).丹参酮ⅡA连续治疗组与单次治疗组比较,其Bcl-2蛋白表达水平更高,Bax蛋白表达水平更低(均P<0.01).与假手术组比较,丹参酮ⅡA连续治疗组的3种性激素水平无明显改变(均P>0.05),而模型组和单次治疗组的FSH、LH水平均明显升高,T水平均明显降低(均P<0.01).结论 丹参酮ⅡA可通过增强Bcl-2基因、抑制Bax基因的表达,从而减轻大鼠睾丸缺血再灌注损伤,并同时参与维持T、FSH和LH三者血清浓度之间的动态平衡.

Abstract

Objective To explore the protective effect and mechanism of tanshinone Ⅱ A on sex hormones and cell apopto-sis of testis following testicular torsion/detorsion in rates.Methods 64 male SD rats were randomly divided into sham operation group,model group,tanshinone Ⅱ A single and continuous treatment groups,with 16 rats in each group.Rats in the single treatment group were injected intraperitoneally with tanshinone Ⅱ A of 25 mg/kg after torsion/detorsion,while the continuous treatment group was maintained with tanshinone Ⅱ A of 25 mg/kg once per day for one week in addition to the above procedures.Rats from each group were killed at 4 and 29 days after reperfusion.The HE and TUNEL staining were used to observe testicular histopathological changes.Western blotting method was used to detect the expression of Bcl-2 and Bax.Sex hormones that Follicle stimulating Hor-mone(FSH),Luteinizing Hormone(LH)and Testosterone(T)were detected by ELISA.Results Compared with model group,less damage of testicular tissue and less germ cell apoptosis were observed in groups which received tanshinone Ⅱ A treatment(all P<0.01),and the improvement was more pronounced in the continuous treatment group than in the single treatment group(P<0.01).The continuous treatment group had higher expression of Bcl-2 and lower expression of Bax,compared with the single treat-ment group(all P<0.01).There were no statistical significantly differences in the levels of sex hormones(FSH,LH,T)between the sham operation group and continuous treatment group(all P>0.05),while there were statistical significantly differences among the sham operation group and model group,single treatment group(P<0.01).Conclusion Tanshinone Ⅱ A can reduce the rat testicular ischemia-reperfusion injury after testicular torsion/detorsion by up regulate the expression of Bcl-2 and down regulate the expression of Bax,as well as maintain a dynamic balance among FSH,LH and T in serum.
