Objective:To establish an improved HPLC method to determine the content of Leucongen.Methods:The isocratic elution HPLC method was used.Thermo ScientificTM C18(4.6 mm×100 mm,3 μm)was adopted with a mobile phase consisting of water-acetonitrile-acetic acid,glacial(75:25:0.3).The column temperature was 30 ℃ and wavelength was 210 nm.The injection volume was 20 μL and flow rate was 1.0 mL/min.Results:The four diastereomers of leucogen can be detected and separated completely by HPLC.There was a good linear relationship in the range of 11.78~294.50 μg/mL.It had good precision and reproducibility.The average recoveries was 98.48%.Conclusion:The method had good specificity and higher sensitivity,resolution,number of theoretical plates,it was suited to determine the content of Leucongen.
