改良HPLC法测定利可君含量

目的:建立一种改良后的HPLC法用于利可君含量测定。方法:采用高效液相色谱法,色谱柱:赛默飞C18(4。6 mm×100mm,3μm),流动相:水-乙腈-冰醋酸(体积比75∶25∶0。3),等度洗脱,柱温:30℃,检测波长:210nm,进样量:20 μL,流速:1。0mL/min。结果:利可君的四个非对映异构体在HPLC中能全部检出且分离彻底,利可君在11。78~294。50 μg/mL范围内线性良好,精密度、重现性优,平均回收率为98。48%。结论:本方法专属性好、灵敏度高、分离度高、理论塔板数高,适用于利可君的含量测定。
Determination of Leucongen by Improved HPLC
Objective:To establish an improved HPLC method to determine the content of Leucongen.Methods:The isocratic elution HPLC method was used.Thermo ScientificTM C18(4.6 mm×100 mm,3 μm)was adopted with a mobile phase consisting of water-acetonitrile-acetic acid,glacial(75:25:0.3).The column temperature was 30 ℃ and wavelength was 210 nm.The injection volume was 20 μL and flow rate was 1.0 mL/min.Results:The four diastereomers of leucogen can be detected and separated completely by HPLC.There was a good linear relationship in the range of 11.78~294.50 μg/mL.It had good precision and reproducibility.The average recoveries was 98.48%.Conclusion:The method had good specificity and higher sensitivity,resolution,number of theoretical plates,it was suited to determine the content of Leucongen.
