Composition Analysis and Antioxidant Activity of Polysaccharide from Reineckia Carnea(Andr.)Kunth
To optimize the extraction process of RCP and to explore the monosaccharide composition and antioxidant activity of RCP in vitro.Using Reineckia carnea(Andr.)Kunth whole grass as raw material,after drying,crushing,degreasing,decolorization and other pre-treatments,on the basis of single factors,orthogonal experiments were used to optimize the extraction process of RCP.HPLC was used to determine the composition and content of monosaccharides,and the antioxidant activity of polysaccharide in vitro was discussed.The best extraction process of RCP was 1∶40(g/mL-1),temperature 100 ℃,time 60 min,and extraction three times,under which the polysaccharide yield was 17.23%±0.78%.HPLC analysis showed that the polysaccharide were mainly composed of eight monosaccharides,namely galacturonic acid 41.90%±0.01%,galactose 9.14%±0.01%,arabinose 4.20%± 0.03%,rhamnose 1.99%±0.02%,glucose 1.98%±0.01%,mannose 1.06%±0.01%,glucuronic acid 0.49%±0.01%.The scavenging rates of DPPH radical,hydroxyl radical,O2-radical and simulated gastric juice NO2-radical were 91.49%±2.76%,29.33%±2.09%,42.37%±2.65%,25.17%±1.06%at a concentration of 1.0 mg/mL,and IC50 values were 0.08,1.67,1.11 and 2.42 mg/mL,respectively.It has certain antioxidant capacity,which is conducive to the development and utilization of polysaccharide in the fields of functional foods,health products and drugs,and provides a theoretical basis for the development and utilization of Reineckia carnea(Andr.)Kunth.
