以二氧化铈-四甲基联苯胺为比色探针的半胱氨酸测定

采用无机物前体自沉淀法制备氧化性二氧化铈纳米簇(CeO2NCs)。3,3′,5,5′-四甲基联苯胺(TMB)被CeO2NCs氧化后生成在652 nm处有特征吸收峰的醌二亚胺(TMB2+),而半胱氨酸(Cys)对TMB2+具有还原作用。基于此原理,建立一种以CeO2NCs-TMB为比色探针的Cys测定方法。常温下,在CeO2NCs质量浓度为1。0 μg/mL,pH值=4。0,反应时间为18 min及TMB浓度为1。2 mmol/L的条件下,当Cys浓度在3。0~50 μmol/L范围内时,TMB2+的吸光度随Cys浓度的升高而线性降低。方法应用于血清样品的测定,加标回收率为99。3%~106。2%。
Determination of Cysteine Using Cerium Dioxide-tetramethylbenzidine as a Colorimetric Probe
Oxidative cerium dioxide nanoclusters(CeO2NCs)were prepared by self-precipitation of inorganic precursor.3,3′,5,5′-tetramethylbenzidine(TMB)is oxidized by CeO2NCs to form quinone diimide(TMB2+)with a characteristic absorption peak at 652 nm,and cysteine(Cys)has a reducing effect on TMB2+.Based on these,a Cys determination method using CeO2 NCs-TMB as a colorimetric probe was established.Under the conditions of room temperature,1.0 μg/mL CeO2NCs,pH value 4.0,reaction time 18 min,and 1.2 mmol/L TMB,the absorbance of TMB2+decreased linearly with the increase of Cys concentration in the range of 3.0~50 μmol/L.This method was applied to determination of Cys in human serum samples with recoveries of 99.3%~106.2%.
