Experimental Study on Oxidative Damage of Liver and Micronucleus Induced by 8-Hydroxyquinoline in Mice
To inestigate the liver oxidative damage and micronucleus formation rate of micronucleus in mice,which treated by different concentrations of 8-hydroxyquinoline.Three dosse groups,positive control(cyclophosphamide),solvent control and blank control were set.After continuous intragastric administration for 7 days,serum alanine aminotransferase(ALT)and aminotransferase(AST)activities were detected.The liver organ coefficient,the content of lipid peroxidation product malondialdehyde(MDA)and the activities of antioxidant indexes catalase(CAT),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)in liver tissue of mice were detected.Results:Compared with the control group,there were no significant differences in liver organ coefficient,ALT,AST activity,MDA content and antioxidant index of male and female mice in solvent group.The activities of SOD enzyme,CAT enzyme and GSH-PX enzyme in 8-hydroxyquinoline group were significantly decreased compared with those in blank group(p<0.05),and the differences in high concentration group were extremely significant(p<0.01).The micronucleus formation rate of male and female mice induced by 8-hydroxyquinoline was significantly higher in high dose group(p<0.05).Conclusion:17.5~70 mg/kg 8-hydroxyquinoline has liver oxidative damage and mutagenic effect of mice.
