高压酸水解-离子交换色谱法测定乳粉中的酵母β-葡聚糖

摘要
目的:采用高压酸水解-离子交换色谱法建立乳粉中酵母β-葡聚糖含量的分析方法.方法:先对乳粉中的酵母β-葡聚糖进行富集,富集得到的酵母β-葡聚糖利用高压酸水解成葡萄糖后利用离子色谱法检测分析,最后通过公式换算计算出酵母β-葡聚糖的含量.结果:葡萄糖在 2~200 mg/L质量浓度范围内的线性关系良好,相关系数r为0.9998.酵母β-葡聚糖的定量限为11.1 mg/100 g.在20,50,100 mg/100 g加标水平下,酵母β-葡聚糖的加标回收率为81.5%~99.8%,相对标准偏差小于5%.结论:该方法简便高效、灵敏度高、准确性好,适用于乳粉中酵母β-葡聚糖的定量检测.
Abstract
Objective:A method for the determination of yeast β-glucan in milk powder was developed by high pressure acid hydrolysis and ion exchange chromatography.Methods:The yeast β-glucan in milk powder was first enriched,and then was hydrolyzed into glucose by high pressure acid,which was analyzed by ion chromatography.The content of yeast β-glucan was calculated by formula conversion.Results:The linear relationship between the peak areas and concentration of glucose in 2~200 mg/L was good,which the correlation coefficient was 0.9998.The quantitative limit of yeast β-glucan was 11.1 mg/100 g.The recoveries of yeast β-glucan at three spiked levels were in the range of 81.5%~99.8%,and the relative standard deviations were all bellow 5%.Conclusion:The study indicated that the method was simplicity,efficiency,high sensitivity and accuracy,which was suitable for the determination of yeast β-glucan in milk powder.
