Optimization of the Simultaneous Extraction Process of Total Flavonoids and Chlorogenic Acid from Honeysuckle Leaves by Response Surface Method
The process of simultaneous extraction of total flavonoids and chlorogenic acid from honeysuckle leaves by pressurized auxiliary solvent was optimized by using the response surface method. On the basis of single factor experiment, the yield of total flavonoids and chlorogenic acid was comprehensively evaluated as the dependent variable, and the liquid-material ratio, extraction pressure, solvent volume fraction, extraction temperature and extraction time were optimized by response surface method. The results showed that the optimized process parameters of pressurized auxiliary solvent synchronous extraction were as follows:liquid-material ratio 26.5 mL/g,extraction pressure 3.1 MPa,solvent volume fraction 60.5%,extraction temperature 90.3 ℃,and extraction time 32 min.Under the conditions of this process, the yield of total flavonoids extracted from honeysuckle leaves was 205.3 mg/g,the yield of chlorogenic acid was 24.5 mg/g,and the relative error between OD value and theoretical predicted value was 1.0%.
