Simultaneous Detection of Multiple Drugs and Their Metabolites in Urine by UPLC-MS
A quantitative internal standard method has been established to detect 14 types of abused drugs and their metabolites in human urine.The analytical instrument is AB SCIEX 6500+triple quadrupole ultra-high performance liquid chromatography tandem mass spectrometer,with a C18 chromatographic column and a mobile phase of 0.1%formic acid aqueous solution and acetonitrile.The ion source mode is ESI positive ion source,and accurate quantification is performed using a multi reaction detection mode.The experimental results showed that the linear relationship of the 14 substances to be tested was very good,with a minimum detection limit of 0.03 ng/mL,high accuracy,and a recovery rate range of 83.46%~114.60%.The precision was good,with a daily precision of less than 10%and a daytime precision of less than 9%.Moreover,the substrate effect of the samples was relatively small after pre-treatment,and the samples remained stable after multiple freeze-thaw cycles.This method is simple and convenient to operate,with accurate qualitative and quantitative results.The experimental results can be replicated and can be used to detect various drugs and their metabolites in the urine of drug users.
