采用超声辅助对火参果中多酚类化合物提取,利用正交实验确定火参果中多酚类化合物的最佳提取条件。在碱性条件下对多酚提取物显色,以没食子酸为对照品,采用紫外吸收光谱法测定火参果中总多酚的含量。结果表明,没食子酸对照品在0。02~0。10 mg/mL质量浓度范围内与吸光度存在良好的线性关系,线性回归方程:Y=10。19X+0。0011,R2=0。999 4。火参果中总多酚最优提取工艺为:超声提取时间50 min、料液比1∶50(g∶mL)、乙醇体积分数60%。
Optimization of Extraction of Polyphenols from Cucumis Metuliferus
The extraction and determination of polyphenols from Cucumis Metuliferus were studied in the paper.Ultrasonic-assisted extraction method was used to extract polyphenols from Cucumis Metuliferus.Orthogonal experimental method was used to confirm the optimal extraction process of polyphenols in Cucumis Metuliferus.The content of the total polyphenols in Cucumis Metuliferus was determined by Ultraviolet-Visible Spectrophotometry with gallic acid as reference substance in an alkaline environment.The absorbance and mass concentration were a good linear relationship at the mass concentration of gallic acid within the range of 0.02~0.10 mg/mL.The regression equation was Y=10.19X+0.001 100,R2=0.999 4.The optimal extraction conditions for polyphenols from Cucumis Metuliferus were extraction time 50 min,material-liquid ratio 1∶50(g∶mL)and ethanol concentration 60%.
