Cloning and Functional Characterization of GbTPS3 Gene in Ginkgo biloba
Objective:Based on deep transcriptome sequencing of Ginkgo biloba,a prokaryotic expression system engineering strain was constructed to systematically study the function of terpenoid synthase in Ginkgo biloba.Methods:Based on the prediction of protein function,different prokaryotic expression systems were used for functional characterization and GC-MS detection analysis.Results:The GbTPS3 gene of Ginkgo biloba can simultaneously express two monoterpenoids(borneol and 2-pinanol).Conclusion:Ginkgo biloba GbTPS3 is Class Ⅰ terpenoid synthase enzymes,mainly responsible for participating in the synthesis reaction of terpenoid compounds.Its characteristic is that the randomness of carbon skeleton rearrangement can produce different carbocation intermediates.In the case of a unique substrate,a single terpenoid synthase often produces multiple products.
