猪毛菜总生物碱提取工艺优化研究

以猪毛菜总生物碱提取率为指标,通过单因素试验和响应面试验优化猪毛菜总生物碱提取工艺。以DPPH和羟基自由基清除率确认猪毛菜总生物碱抗氧化能力。结果表明,猪毛菜总生物碱最优提取工艺为液料比36∶1(mL∶g)、超声提取时间60 min、复合酶含量2%、酶解时间1。4 h、乙醇体积分数57%,在最优条件下,猪毛菜总生物碱提取率可达0。698 2%。抗氧化活性实验表明,随着猪毛菜生物碱浓度增加,抗氧活性随之增加,当猪毛菜生物碱质量浓度为0。5 mg/mL时,DPPH自由基和羟基自由基清除率分别达到84。34%和75。69%。本研究优化了猪毛菜总生物碱的提取工艺,初步分析猪毛菜生物碱抗氧化活性,为猪毛菜有效药用成分的分析研究和功能性食品的开发提供科学依据。
Studies on Optimization of Extraction Process for Total Alkaloids from Salsola collina Pall
The extraction rate total alkaloids from Salsola collina was used as the indicator,the extraction process of total alkaloids from S.collin was optimized through single factor test and response surface methodology,and DPPH and hydroxyl radical clearance was confirmed the antioxidant capacity of S.collin total alkaloids.The results showed that the optimal extraction process conditions for S.collin total alkaloids were respectively the liquid-material ratio of 36∶1(mL∶g),ultrasonic extraction time 60 min,compound enzyme addition 2%,enzymolysis time 1.4 h,ethanol volume fraction 57%.Under this condition,the extraction rate of the total alkaloids can reach 0.698 2%.The antioxidant activity experiment showed that the antioxidant activity increased with the concentration of alkaloids,when the S.collin total alkaloids concentration of 0.5 mg/mL,the DPPH radical and hydroxyl radical clearance reached 84.34%and 75.69%,respectively.This study optimized the extraction process of the total alkaloids in Salsola collina,preliminarily analyzed the antioxidant activity of the alkaloids,and provided a scientific basis for the analysis of the effective medicinal components of Salsola collina and the development of functional food.
