目的:建立HPLC法测定富马酸贝达喹啉中的SR-异构体.方法:采用高效液相色谱法手性色谱柱Phenomemex Lux Cellulose-1(250 mm×4.6 mm,5 µm),以0.05 mol/L醋酸铵溶液(pH值7.0)-甲醇(V醋酸铵溶液∶V甲醇=40∶60)为流动相,检测波长为226nm,柱温为25℃,流速为0.5 mL/min.结果:富马酸贝达喹啉与SR-异构体能得到良好的分离,出峰时间合适,峰面积与浓度在一定范围内呈良好的线性关系.结论:本测定方法准确可靠、专属性强、灵敏度高、重复性好,适用于富马酸贝达喹啉中SR-异构体的测定.
Determination of SR-enantiomers in Bedaquiline Fumarate
Objective:To establish a HPLC method for determination of SR-enantomer in bedaquiline fumarate.Methods:The method was determined by HPLC with Phenomenex Lux Cellulose-1(250 mm×4.6 mm,5 μm)chiral column.The mobile phase was ammonium acetate buffer(pH value 7.0)-methanol(40∶60,volume ratio),detection wavelength was 226 nm.The column temperature was 25 ℃.The flow rate was 0.5 mL/min.Results:The SR-enantiomer could be well separated from the main peak of bedaquiline fumarate with a suitable retention time,and the calibration curves showed a good linear relationship within a certain range.Conclusion:The method is accurate and reliable,has strong specificity,high sensitivity and good repeatability,and is suitable for the determination of SR-enantomer in bedaquiline fumarate.
