黄蜀葵花药材总多糖含量测定方法研究

目的:建立黄蜀葵花药材中总多糖的含量测定方法,为其质量控制提供依据.方法:以葡萄糖为对照,用苯酚-硫酸法对样品显色,采用紫外-可见分光光度法在485 nm波长处测定总多糖的含量,并对方法学进行研究.结果:总多糖在 2.270~13.620 μg/mL范围内呈良好线性关系;其回归方程为Y=0.061 218 6X-0.017 805 8(r=0.999),加样回收率为99.15%,RSD为0.66%(n=9).结论:采用苯酚-硫酸法测定黄蜀葵花药材中总多糖的含量,方法稳定,精密度高,重复性好,适用于黄蜀葵花药材的多糖含量测定.
Study on the Determination of Polysaccharide Content in Abelmoschi Corolla
Objective:To establish the method to deteminate the content of polysaccharide in Abelmoschi Corolla,it may provide the basis for quality control of Abelmoschi Corolla.Methods:Phenol-sulfu-ricacid method was used for chromogenic recction.The content of total polysaccharide was measured by UV spectrophotometry at 485 nm.Results:The total polysaccharides calibration curve was at the range of 2.270~13.620 μg/mL,with regression function being Y=0.061 218 6X-0.017 805 8(r=0.999).The recowery of calycosin was 99.15%,RSD为 0.66%(n=9).Conclusion:The phenolsulfuric acid method is stable,reliable,simple and easy to operate in the determination of polysaccharides in Abelmoschi Corolla.
