黔产杠板归中总黄酮提取工艺的研究

目的:对黔产杠板归中有效成分总黄酮的提取工艺进行研究,寻找其最佳的提取工艺。方法:采用乙醇热回流法与超声提取法两种常规提取方法进行提取,选出提取率较高的工艺,再结合酶解法再次进行提取,通过单因素实验获得最佳酶解条件,再结合正交试验优选出最佳组合。结果:最佳提取工艺为超声提取法辅助酶解法,最佳提取条件为纤维素酶质量浓度、酶解温度、酶解pH值、酶解时间分别0。3 mg·mL-1、50℃、6、100 min。结论:通过超声提取法辅助酶解法对黔产杠板归中总黄酮进行提取,工艺操作简单、重现性高、提取率较高,可用于黔产杠板归中总黄酮的大规模提取。
Study on the Extraction Technology of Total Flavones from the Centralization of Guizhou Polygonum Perfoliatum L
Objective:The extraction technology of total flavonoids from Guizhou produced Polygonum perfoliatum L was studied,and the best extraction technology was found.Method:Two conventional extraction methods,ethanol thermal reflux method and ultrasonic extraction method,were used for extraction.The process with higher extraction rate was selected,and then combined with enzymatic hydrolysis to extract again.Combined with enzymatic hydrolysis method,the optimal enzymatic conditions were obtained by single factor experiment,and the optimal combination was optimized by orthogonal test.Result:The optimal extraction process isultrasound assisted enzymatic hydrolysis.The optimal extraction conditions are enzymatic hydrolysis temperature of 50 ℃,enzymatic hydrolysis pH of 6,enzymatic hydrolysis time of 100 minutes,and cellulase concentration of 0.3 mg·mL-1.Conclusion:The total flavonoids from Guizhou produced Polygonum perfoliatum L were extracted by ultrasonic-assisted enzymatic method.This process is simple,highly reproducible,and has a high extraction rate,and can be used for largescale extraction of total flavonoids in Polygonum perfoliatum L from Guizhou province.
