超高效液相色谱—串联质谱测定毛发中依托咪酯等10种精神活性物质

建立一种超高效液相色谱—串联质谱(UPLC-MS/MS)同时测定毛发中依托咪酯等10种精神活性物质的方法。采用快速便捷的湿法冷冻研磨并结合超声提取毛发中10种精神活性物质,通过Kinetic Biphenyl(100 mmx2。1 mm,1。7 μm)色谱柱分离,在电喷雾离子源正离子模式(ESI+)下,多反应监控(MRM)模式检测。结果表明,毛发中依托咪酯等10种精神活性物质检出限在0。001~0。006 ng/mg,定量限在0。003~0。001 9 ng/mg;在0。02~2 ng/mg范围内线性关系良好,R2均大于0。994 3;在低、中、高(0。02,0。2,2 ng/mg)三种添加浓度中基质效应在84。53%~112。01%,提取回收率在88。68%~112。88%,日内、日间精密度RSD分别在1。48%~6。32%和1。25%~7。85%。本方法专属性好、灵敏度高、准确性好,可用于快速准确测定毛发中依托咪酯等10种精神活性物质。
Determination of 10 Psychoactive Substances in Hair by UPLC-MS/MS
An ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)method was developed for the simultaneous determination of 10 psychoactive substances in hair.A rapid and convenient wet freezing grinding method combined with ultrasonic extraction was used to extract 10 psychoactive substances in hair.The samples were separated by Kinetic Biphenyl(100 mm×2.1 mm,1.7 μm)and detected by multiple reaction monitoring(MRM)mode under positive ion mode(ESI+)of electrospray ion source.The results showed that the detection limits of 10 psychotropic substances in hair were between 0.001~0.006 ng/mg,and the quantitative limits were between 0.003~0.001 9 ng/mg.The linear relationship was good in the range of 0.02~2 ng/mg,and R2 was greater than 0.994 3.In the low,medium and high(0.02,0.2,2 ng/mg)concentrations,the matrix effect was between 84.53%and 112.01%,the extraction recovery was between 88.68%and 112.88%,and the intra-day and inter-day precision RSD were 1.48%~6.32%and 1.25%~7.85%,respectively.The method has good specificity,high sensitivity and good accuracy,and can be used for rapid and accurate determination of 10 psychoactive substances in hair.
