摘要

目的 建立山药块茎中山药素含量的超高效液相色谱串联质谱(UPLC-MS/MS)检测方法.方法 样品经70%甲醇提取溶剂超声提取,离心,用0.22 μm的微孔滤膜过滤,以Shim-pack Velox C18色谱柱进行化合物的分离,梯度洗脱,采用电喷雾离子源(ESI+)、多反应监测(MRM)模式进行分析.结果 3种山药素分别在0.500~20.000,0.050~5.000,0.100~2.000 mg/L浓度范围内与峰面积呈良好的线性关系,相关系数均>0.99;山药样品平均加标回收率为85%~101%,RSD为1.20%~4.74%.结论 经方法学验证,该方法前处理简单,检测快速、灵敏高效,可用于山药中山药素含量定量检测分析,为山药的大面积种植、产品开发、质量控制与评价提供技术支撑.

Abstract

Objective To establish an ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)method for the determination of batatasin compounds in Dioscoreae Bhizoma.Methods The samples were ultrasonically extracted with 70%methanol extraction solvent,followed by high-speed centrifugation,then were filtered with a 0.22 μm microporous membrane.Chromatographic separation was carried out on Shim-pack Velox C18 column with gradient elution.Electrospray ionization(ESI+)and multi-reaction monitoring(MRM)were used for analysis.Results Three kinds of batatasin showed good linear relationgships in the ranges of 0.500-20.000,0.050-5.000,0.100-2.000 mg/L,respectively.The average recoveries of batatasin were 85%-101%,with RSDs of 1.20%-4.74%,respectively.Conclusion The method is sensitive and accurate.It can be used for quantitative detection and analysis of batatasin content.It provides technical support for large-scale planting,product development,quality control and evaluation of Dioscoreae Rhizoma.
