摘要

目的 建立氨基酸分析仪测定注射用透明质酸钠复合溶液中脯氨酸、甘氨酸和丙氨酸含量的方法.方法 供试品中的氨基酸经阳离子交换树脂柱分离,于135 ℃下与茚三酮进行柱后衍生化显色反应,脯氨酸的检测波长为440 nm,其他氨基酸的检测波长为570 nm,外标法计算各氨基酸含量.结果 脯氨酸、甘氨酸和丙氨酸线性范围分别为0.0148~0.074,0.0207~0.103,0.0247~0.1235 mg/ml(r2=0.9998);平均回收率分别为98.2%,98.5%和98.3%,RSD分别为0.55%,0.22%和1.40%;定量限分别为13.28,0.828,0.988μg.结论 方法重现性好、灵敏度高、检测结果准确且操作简单,适用于注射用透明质酸钠复合溶液中氨基酸含量的测定.

Abstract

Objective To establish a method for the determination of proline,glycine and alanine in injectable compound solution of hyaluronic acid by amino acid autoanalyzer.Methods The amino acids in the samples were separated by cation exchange chromatographic column,and reacted with ninhydrin by post-column derivatization at 135 ℃.The detection wavelength of proline was 440 nm,and that of other amino acids was 570 nm.The amino acids were quantified by external standard method.Results Proline,glycine and alanine showed good linear relationships in the range of 0.0148-0.074,0.0207-0.103 and 0.0247-0.1235 mg/ml,respectively(r2=0.9998).The recoveries were 98.2%,98.5%and 98.3%,and RSDs were 0.55%,0.22%and 1.40%;the limit of quantifications were 13.28,0.828 and 0.988 μg,respectively.Conclusion This method is repeatable,sensitive,accurate and simple,suitable for the quantitative analysis of amino acids in injectable compound solution of hyaluronic acid.
