Study on GC-MS Chromatographic Fingerprint of Volatile Oil from Ferula krylovii Korov and Its Anti-cervical Cancer Effect
Objective To establish gas chromatography-mass spectrometry(GC-MS)fingerprint of volatile oil from Ferula krylovii Korov and investigate its anti-cervical cancer effect.Methods The volatile oil from Ferula krylovii Korov was extracted by organic solvent extraction,and the components were identified by GC-MS.MTT colorimetric assay was used to investigate the inhibitory effect of different concentrations of volatile oil from Ferula krylovii Korov on the proliferation of cervical cancer Hela cells.Effect of the volatile oil on the migration and invasion ability of cervical cancer Hela cells was detected by Transwell method.The apoptosis rate of Hela cells under the effect of the volatile oil from Ferula krylovii Korov was detected by flow cytometry.Results 58 kinds of chemical constituents were identified in the volatile oil of Ferula krylovii Korov,most of which were progesterone.The volatile oil of Ferula krylovii Korov showed a certain inhibitory effect on the proliferation of human cervical cancer Hela cells with IC50 of 55.21 μg/ml.Compared with the control group,the number of the migrating and invading cells treated with the high and low doses of the volatile oil from Ferula krylovii Korov significantly reduced(P<0.01).Conclusion The compounds with highest content in volatile oil from Ferula krylovii Korov are ketones.The extract of Ferula krylovii Korov has certain anti-tumor effect.
