Rapid Determination of Chloramphenicol,Thiamphenicol and Florfenicol Residues in Mutton by QuEChERS-UPLC-MS/MS
Objective To establish an analytical method for the determination of chloramphenicol, thiamphenicol and florfenicol residues in mutton by QuEChERS-ultra performance liquid chromatography-tandem triple quadrupole mass spectrometry(QuEChERS-UPLC-MS/MS).Methods The samples were homogenized by a grinder and extracted with acetonitrile. The extract was purified by QuEChERS-1010 and QuEChERS-0101,determined by UPLC-MS/MS and quantified by internal standard method. For detection, electrospray negative ion scanning and multiple reaction monitoring were adopted. Results Chloramphenicol, thiamphenicol and florfenicol showed good linear relationships in the range of 4-50 μg/L,with correlation coefficients above 0.998.At the concentrations of 5.0,10.0,30.0 μg/kg,the recoveries of three drugs were in the range of 93.17%-109.91%,and the relative standard deviations(RSDs)ranged from 2.33%to 6.14%.The limits of detection of the method were 0.01-0.03 μg/kg,and the limits of quantification were 0.02-0.09 μg/kg.Conclusion This method is simple, rapid, accurate and sensitive, and meets the detection requirements for the determination of chloramphenicol, thiamphenicol and florfenicol residues in mutton.
