摘要
目的 制备相对分子质量(Mr)不同的核桃肽,并初步探究核桃肽经Cy7标记后在小鼠体内分布情况.方法 利用不同酶降解核桃粕制备核桃肽,GPC色谱法测定核桃肽Mr,利用Cy7标记的核桃肽进行小鼠活体成像,观察其在小鼠体内的分布情况.结果 不同酶酶解得到的样品Mr占比不同,样品Ⅰ小分子量成分占比高,收率高,通过Cy7标记不同样品在小鼠体内呈现不同的荧光强度,Cy7-样品Ⅰ在脑部显现荧光信号,Cy7-样品Ⅱ和Cy7-样品Ⅲ仅在接受注射的尾部显现强荧光信号.结论 不同酶制备的核桃肽在小鼠体内呈现不同的分布,样品Ⅰ中可能含有增强脑部荧光的肽段.
Abstract
Objective To prepare walnut peptides with different molecular weights and preliminarily investigate the distribution of walnut peptides in mice after being labeled with Cy7.Methods Walnut peptides were prepared by degradation of walnut dregs with different enzymes,the relative molecular weight(Mr)of walnut peptides was determined by GPC chromatography,and the Cy7-labeled walnut peptides were used for in vivo imaging of mice to observe their distribution in mice.Results The Mr of samples obtained by different enzymes was different.Sample Ⅰ had a high proportion of small molecular weight components and a high yield.Different samples were labeled by Cy7 to show different fluorescence intensities in mice.Cy7-sample Ⅰ showed fluorescence signals in the brain,and Cy7-sample Ⅱ and Cy7-sample Ⅲ only showed strong fluorescence signals in the tail of the injection.Conclusion Walnut peptides prepared by different enzymes have different distributions in mice,and sample Ⅰ may contain peptides which enhance brain fluorescence.
