目的 建立LC-MS/MS测定桃红四物汤中芍药苷含量的方法.方法 色谱分离采用Agilent Eclipse Plus C18(2.1 mm×50 mm,5 μm)色谱柱,流动相为甲醇和水(含0.1%甲酸),梯度洗脱.质谱采集选用ESI负离子及多反应监测模式.结果 芍药苷在0.1~50 μg/ml范围内线性关系良好,专属性、回收率、精密度、均一性均符合要求.结论 本法专属性强、灵敏度高、分析时间短,可作为桃红四物汤中芍药苷的含量测定.
Determination of Paeoniflorin in Taohong Siwu Decoction by LC-MS/MS
Objective To establish a method for the determination of paeoniflorin in Taohong Siwu Decoction by LC-MS/MS.Methods The separation was achieved on an Agilent Eclipse Plus C18(5 μm,2.1 mm×50 mm)column using a gradient mobile phase system of methanol-water(containing 0.1%formic acid).ESI negative ionization and multiple reaction monitoring mode(MRM)were used for mass spectrum acquisition.Results The linear range of paeoniflorin was 0.1 to 50 μg/ml.Specificity,recovery,precision and uniformity all met the requirements.Conclusion This method has strong specificity,high sensitivity,short analysis time,and can be used for the determination of paeoniflorin in Taohong Siwu Decoction.
