摘要

目的 建立高效液相色谱-荧光检测器法同时测定乳及乳制品中维生素A和维生素E的分析方法.方法 样品经水解皂化、液液萃取、蒸发浓缩后,选用PFP色谱柱分离,以甲醇-水(9:1,V/V)等度洗脱,高效液相色谱-荧光检测器检测,外标法定量,计算维生素A、维生素E的含量.结果 维生素A和4种维生素E异构体在0.10~5.00 μg/ml范围内呈现良好线性,方法检出限为0.0100 μg/g、方法定量限为0.0400 μg/g,维生素A的平均回收率在85.0%~97.0%之间,RSD≤6.41%(n=6);4种维生素E异构体的平均回收率在84.0%~101%之间,RSD≤7.04%(n=6).结论 本方法简便快速,灵敏准确,可满足乳及乳制品中维生素A和维生素E的同时测定.

Abstract

Objective To establish a method for simultaneous determination of vitamin A(VA)and vitamin E(VE)in milk and dairy products by high performance liquid chromatography with fluorescence detector(HPLC-FLD).Methods The samples were hydrolyzed and saponified,liquid-liquid extracted,evaporated and concentrated,then were separated on a PFP column,using methanol-water(9:1,V/V)as mobile phase of isocratic elution,and were detected by HPLC-FLD.External standard method was adopted for quantification.Results The standard curves of VA and 4 kinds of VE all exhibited ideal linear relation over the concentration range of 0.10-5.00 μg/ml,the limit of detection was 0.0100 μg/g,and the limit of quantification was 0.0400 μg/g.The average recoveries of VA and 4 kinds of VE were 85.0%-97.0%with RSDs<6.41%(n=6)and 84.0%-101%with RSDs<7.04%(n=6),respectively.Conclusion The method is simple,rapid,sensitive and accurate,which can be used for simultaneous determination of VA and VE in milk and dairy products.
