Research on Mechanism of Xiaoyao Powder in Treatment of Chloasma Based on Network Pharmacology and Experimental Validation
Objective To analyze the mechanism of Xiaoyao Powder in the treatment of chloasma based on network pharmacology and experimental validation.Methods The bioactive components and related targets of Xiaoyao Powder were retrieved from TCMSP and other databases and related literature,and the network of drug-active components-targets of Xiaoyao Powder was constructed.Based on the databases such as Drugbank and Genecards,the disease targets related to chloasma were established and matched with the effective component targets of Xiaoyao Powder to obtain potential therapeutic targets.GO bioprocesses,KEGG signaling pathways,and protein-protein interaction(PPI)analyses of potential therapeutic targets were performed based on Metascape and DAVID databases.The topology analysis of PPI network was performed by Cytoscape software.Then the above results were verified by experiments.Results There were 928 effective chemical components in Xiaoyao Powder,including 85 kinds from Paeoniae Radix Alba,55 kinds from Atractylodis Macrocephalae Rhizoma,349 kinds from Bupleuri Radix,125 kinds from Angelicae Sinensis Radix,34 kinds from Poria,280 kinds from Glycyrrhizae Radix et Rhizoma,and 155 target genes of the active components were also obtained.Among the top 10 important active ingredients in the drug-active ingredient-target network,there were 4 ingredients from Bupleuri Radix,3 ingredients from Paeoniae Radix Alba,1 ingredient from Atractylodis Macrocephalae Rhizoma and 2 ingredients from Glycyrrhizae Radix et Rhizoma.A total of 49 chloasma related targets were collected,including 24 potential target genes of Xiaoyao Powder in the treatment of chloasma.KEGG enrichment analysis showed that among all the 20 signaling pathways,top 10 signaling pathways were obtained.GO function enrichment analysis showed that the top 10 biological processes included molecular regulatory function,phosphorylation,etc,the top 10 molecular functions include cell delivery parts,biofilms,etc,and the top 10 cell locations included nucleotides,purine nucleotides,etc.A core network of 36 nodes was obtained after the extraction of PPI network,among which HSP90AA1,HSP90AB1,NTRK1,CUL3,APP,EGFR,TP53,UBC,MCM2 and PRKACA genes were the core targets.Experimental verification proved that Xiaoyao Powder could significantly improve the chloasma condition of rats.It could increase serum SOD level,decrease serum Tyr and MDA contents,and improve the skin tissue structure and skin HMB45 expression at chloasma.Conclusion Xiaoyao Powder can treat chloasma through multiple components,multiple targets and multiple pathways.The effective ingredients for treatment are mainly from Bupleuri Radix,Paeoniae Radix Alba,Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix et Rhizoma,while the core targets are mainly HSP90AA1 and other genes.
