HPLC波长切换法同时测定畅中丸中6种主成分的含量

目的 建立高效液相色谱法同时测定畅中丸中栀子苷、芍药苷、柚皮苷、橙皮苷、新橙皮苷、盐酸小檗碱6种指标性成分的含量.方法 以Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm)为色谱柱,以乙腈(A)-0.1%磷酸(B)作为流动相,进行梯度洗脱;检测波长为238,230,284,345 nm;流速为1.0 ml/min、柱温为30℃.结果 栀子苷、芍药苷、柚皮苷、橙皮苷、新橙皮苷、盐酸小檗碱在各自的测定范围内,线性关系良好,相关系数(r)均大于0.999,平均回收率在97.98%~99.53%之间,RSD均小于2.0%.3批次畅中丸中栀子苷、芍药苷、柚皮苷、橙皮苷、新橙皮苷、盐酸小檗碱含量分别为2.307~2.395,1.806~1.895,11.392~11.959,0.507~0.544,5.314~5.433,2.406~2.497 mg/g.结论 所建立的测定方法简便,准确灵敏,重复性较好,可用于畅中丸中6种指标性成分的质量控制.
Simultaneous Determination of Six Index Components in Changzhong Pills by HPLC-Wavelength Switching Method
Objective To establish an HPLC method for simultaneous determination of six index components including geniposide,paeoniflorin,naringin,hesperidin,neohesperidin and berberine hydrochloeide in Changzhong Pills.Methods The chromatography was performed on Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm)column with acetonitrile(A)-0.1%phosphoric acid(B)as mobile phases for gradient elution.The detection wavelengths were set at 238,230,284,345 nm,respectively.The flow rate was 1.0 ml/min and the column temperature was 30℃.Results The linear relationships of geniposide,paeoniflorin,naringin,hesperidin,neohesperidin and berberine hydrochloeide were good within their respective determination ranges.The correlation coefficients(r)were greater than 0.999,the average recoveries were between 97.98%-99.53%,with RSDs less than 2.0%.The contents of geniposide,paeoniflorin,naringin,hesperidin,neohesperidin and berberine hydrochloeide in three batches of Changzhong Pills were 2.307-2.395,1.806-1.895,11.392-11.959,0.507-0.544,5.314-5.433,2.406-2.497 mg/g,respectively.Conclusion The established method is simple,accurate,sensitive and reproducible,and can be used for the quality control of 6 index components in Changzhong Pills.
