Simultaneous Determination of Four Components in Codonopsis Radix by HPLC
Objective To establish an HPLC method for simultaneous determination of tangshenoside Ⅰ,lobetyolinin,lobetyolin and atractylenolide Ⅲ in Codonopsis Radix.Methods HPLC was performed on V-0203052-5250 Svea(4.6 mm250 mm,5 µm)column with 0.1%phosphoric acid solution-acetonitrile as mobile phases for gradient elution.The detection wavelengths were set at 220 nm(for detection of tangshenoside Ⅰ and atractylenolide Ⅲ)and 210 nm(for dection of lobetyolinin and lobetyolin),respectively.The flow rate was 0.8 ml/min.The column temperature was 25℃.Results The linear relationships of tangshenoside Ⅰ,lobetyolinin,lobetyolin and atractylenolide Ⅲ were good(r2≥0.9999,n=6)within their ranges of 3.13-200.00,12.50-400.00,6.25-200.00,0.39-12.50 µg/ml,respectively.The average recoveries were 82.20%-98.94%.Conclusion The method is of high repeatability and good accuracy,and can be used for quality control of Codonopsis Radix and its products.
