Determination of Four Tannins in Fruits and Kernels of Chebulae Fructus by HPLC
Objective To establish a method for simultaneous determination of four tannins(corilagin,chebulagic acid,chebulinic acid and ellagic acid)in the fruits and kernels of Chebulae Fructus.Methods Agilent Zorbax C18 column(250 mm×4.6 mm,5 μm)was used,the mobile phase was methanol(A)-0.1%phosphoric acid(B),and the gradient elution was performed at a flow rate of 1.0 ml/min.The detection wavelength was set at 270 nm,and the column temperature was 25 ℃.Results The four components in the fruits and kernels of Chebulae Fructus were well separated.Corilagin,chebulagic acid,chebulinic acid,and ellagic acid showed good linear relationships in the ranges of 0.0813-5.9936,0.0311-11.4822,0.3271-4.9008 and 0.0760-1.4935 μg,respectively.The linear equations were Y=1915X+336.9,Y=1532.1X+554.89,Y=2077.9X-116.96 and Y=7194.4X-53.568,respectively.The results of precision and repeatability tests were good,with RSDs all less than 5%.The test solution was stable within 12 h at room temperature.The average recoveries of four tannins in fruits and kernels were 99.73%-102.54%and 99.34%-103.32%,respectively,with RSDs of 0.10%-0.86%and 0.93%-1.97%,respectively.The difference between the contents of chebulagic acid in fruits and kernels was the most significant.Conclusion The method is simple,accurate and reproducible.It can be used for the simultaneous determination of corilagin,chebulagic acid,chebularic acid and ellagic acid in the fruits and kernels of Chebulae Fructus.
