安神定志灵对ADHD模型鼠血清多巴胺及S100β蛋白的影响

摘要
目的:观察安神定志灵对ADHD模型鼠(SHR)血清单胺类神经递质多巴胺(DA)及神经营养因子S100β蛋白(S100β蛋白)的影响.方法:将30只SHR大鼠随机分为模型组、利他林组及安神定志灵低剂量组、中剂量组、高剂量组,将6只同龄雄性Wistar京都大鼠(WKY)设为对照组,各组灌胃给药2周后取血清,采用ELISA法及BCA蛋白定量法分别检测血清DA及S100β蛋白含量.结果:模型组大鼠血清DA、S100β蛋白含量较正常组明显降低(P<0.05);安神定志灵各剂量组及利他林组大鼠血清DA、S100β蛋白含量均较模型组显著升高(P<0.05).结论:DA和S100β蛋白可能是安神定志灵治疗ADHD的有效靶点之一.
Abstract
Objective:To explore the effects of Anshen Dingzhiling pills(ASDZLP) on serum dopamine(DA) and neurotrophic factor protein S100β(protein S100P) of spontaneously hypertensive rats(SHR) with attention deficit hy-peractivity disorder (ADHD). Methods: Total 30 SHR rats were randomly divided into five groups; the model group, methylphenidate group (treated by litalin), low-dose ASDZLP group, medium-dose ASDZLP group, and high-dose ASDZLP group. There were 6 rats in each group. Meanwhile,6 WKY rats were set as the normal control group. After given drugs two weeks,the contents of serum DA and protein SlOOp were detected by ELISA and BCA methods. Results;Compared with normal control group,serum levels of DA and protein SlOOp of model group were significantly decreased (P<0.05). Compared with model group,serum levels of DA and protein SlOOp of the methylphenidate group and ASDZLP groups were increased(P<0.05). Conclusion;The DA and the protein SlOOp maybe the effective targets of Anshen Dingzhiling pills in treating ADHD.
