复方秦柏洗剂及土槐丹四物汤对银屑病模型小鼠炎症因子的影响

目的:观察复方秦柏洗剂及土槐丹四物汤对小鼠耳肿胀炎症及鼠尾鳞片表皮颗粒层的影响,为银屑病的治疗提供依据.方法:选取雄性KM小鼠48只,按照随机数字表法分为空白组、模型组、卡泊三醇软膏组、复方秦柏洗剂组、土槐丹四物汤组、复方秦柏洗剂+土槐丹四物汤组,每组各8只.空白组、模型组予生理盐水涂搽加灌胃,各给药组分别给予各自药物涂搽加灌胃,连续7 d,末次给药1 h后除空白组外在其余各组小鼠右耳耳郭正反两面均匀涂抹20 μL二甲苯以建立耳肿胀炎症模型.观察耳肿胀程度;酶联免疫吸附试验(ELISA)法测定血清中白细胞介素-17(IL-17)、白细胞介素-23(IL-23)含量;苏木精-伊红(HE)染色观察耳部皮肤皮损病理学改变;免疫组化测定耳组织中γ干扰素(IFN-γ)、白细胞介素-6(IL-6)的表达情况.选取雌性KM小鼠40只,按照随机数字表法分为空白组、卡泊三醇软膏组、复方秦柏洗剂组、土槐丹四物汤组、复方秦柏洗剂+土槐丹四物汤组,每组各8只,直接利用鼠尾鳞片表皮模型,空白组予生理盐水涂搽加灌胃,各给药组分别给予各自药物涂搽加灌胃,连续14 d.测定鼠尾鳞片表皮颗粒层细胞数量;ELISA法测定血清中IFN-γ、IL-6的含量;HE染色观察鼠尾鳞片表皮颗粒层细胞形成情况;免疫组化测定尾部皮肤组织中IL-17、IL-23的表达情况.结果:与模型组比较,各治疗组雄鼠的耳肿胀程度均减轻(P<0.01),雌鼠尾鳞片表皮颗粒层细胞均增多(P<0.01);复方秦柏洗剂+土槐丹四物汤组雄鼠血清中IL-17、IL-23和雌鼠血清中IFN-γ、IL-6的含量显著下降(P<0.01);雄鼠耳组织中IFN-γ、IL-6和雌鼠尾部皮肤组织中IL-17、IL-23的表达也显著下降(P<0.01).与卡泊三醇软膏组比较,复方秦柏洗剂+土槐丹四物汤组上述指标差异无统计学意义(P>0.05).结论:复方秦柏洗剂联合土槐丹四物汤可通过降低炎症因子IL-17、IL-23、IFN-γ、IL-6的表达抑制炎症和促进表皮正常分化,从而起到治疗银屑病的作用.
Effect of Compound Qinbai Lotion(复方秦柏洗剂)and Tuhuaidan Siwu Decoction(土槐丹四物汤)on Inflammatory Factors in Psoriasis Model Mice
Objective:To observe the effect of Com-pound Qinbai Lotion(复方秦柏洗剂)and Tuhuaidan Siwu Decoction(土槐丹四物汤)on ear swelling inflammation and the granular layer of tail scale epidermis in mice,to provide the basis for the treatment of psoriasis.Methods:Total 48 male KM mice were selected and divided into the blank group,the model group,the calcipotriol ointment group,Compound Qinbai Lotion group,Tuhuaidan Siwu Decoction group,Compound Qinbai Lotion+Tuhuaidan Siwu Decoction group according to the random number table method,with 8 mice in each group.The blank and model groups were treated with saline topically and by gavaged,while the medication groups were treated with their respective drugs topically and by gavaged for 7 consecutive days.One hour after the last administration,20 µL xylene was applied evenly on both sides of the right ear of mice in other groups except the blank group to establish the ear swelling and inflammation model.The degree of ear swelling was then observed.Enzyme linked immunosorbent assay(ELISA)was used to determine the content of interleukin-17(IL-17)and interleukin-23(IL-23)in the serum.Hematoxylin-Eosin(HE)staining was used to observe the pathological changes in ear skin lesions.Immunohistochemistry was used to measure the expressions of interferon-γ(IFN-γ)and interleukin-6(IL-6)in ear tissue.A total of 40 female KM mice were selected and divided into the blank group,calcipotriol ointment group,Compound Qinbai Lotion group,Tuhuaidan Siwu Decoction group,and Compound Qinbai Lotion+Tuhuaidan Siwu Decoction group according to the random number table method,with 8 mice in each group,using the tail scale epidermis model directly.The blank group was treated with saline topically and by gavaged,while the medication groups were treated with their respective drugs topically and by gavaged for 14 consecutive days.The number of the granular layer of tail scale epidermis in mice was measured.ELISA was used to determine the content of IFN-γ and IL-6 in the serum.HE staining was used to observe the formation of the granular layer of tail scaleepid ermis in mice.Immunohistochemistry was used to measure the expressions of IL-17 and IL-23 in tail skin tissue.Results:Compared with the model group,the degree of ear swelling in the male mice of each treatment group reduced(P<0.01),and the cell number of granular layer of tail scale epidermis in female mice increased(P<0.01).The serum content of IL-17,IL-23 in the male mice and IFN-γ,IL-6 in the female mice of the Compound Qinbai Lotion+Tuhuaidan Siwu Decoction group significantly decreased(P<0.01).The expressions of IFN-y and IL-6 in the ear tissue of male mice and IL-17,IL-23 in the tail skin tissue of female mice also significantly decreased(P<0.01).Compared with the calcipotriol ointment group,there was no statistical significance in the above indicators in the Compound Qinbai Lotion+Tuhuaidan Siwu Decoction group(P>0.05).Conclusions:Compound Qinbai Lotion combined with Tuhuaidan Siwu Decoction can inhibit inflammation and promote normal epidermal differentiation by reducing the expressions of inflammatory factors IL-17,IL-23,IFN-γ and IL-6,thereby playing a role in the treatment of psoriasis.
