Objective:To construct a rapid evaluation model for the study of anti-hypoxia active components in Chinese medicine using model animal zebrafish.Methods:The AB strain zebrafish were used in the experiment,with Hongjingtian Capsule(红景天胶囊)as the positive drug and sodium bisulfite as the hypoxia agent.Based on the neurobehavioral evaluation indicators after hypoxia in zebrafish,the starting time for drug pretreatment and the 24 h,48 h,and 72 h drug pre-protection time were investigated in zebrafish larvae after fertilization which developed to 3d,4d,5d to establish a zebrafish larval hypoxia model.Based on the optimized pretreatment time and pre-protection time,the average heart rate after hypoxia and the number of brain tissue cell apoptosis in each group of zebrafish were detected to validate the hypoxia model.Results:Zebrafish developed until 5 d were selected for the experiments,with the drug pre-protection time of 48 h,and the hypoxia model was established using neurobehavioral evaluation indicators at different stages after hypoxia in zebrafish.Further model evaluation found that the Hongjingtian Capsule pre-protection group significantly enhanced the average heart rate in zebrafish caused by hypoxia and simultaneously reduced the number of brain tissue cell apoptosis caused by hypoxia(P<0.01).Conclusions:The neurobehavioral evaluation of zebrafish at different stages after sodium bisulfite modelling can rapidly evaluate the anti-hypoxia activity model of Chinese medicine samples,providing a key model for the screening and discovery of pharmacodynamic components of Chinese medicine.
