Effect and Kinetics of Different Solvent Extracts of Zhijuzi(Hovenia Dulcis)on Xanthine Oxidase
Objective:To investigate the effect and kinetic model of Zhijuzi(Hovenia Dulcis)extracted by different solvents on xanthine oxidase(XOD),and to provide experimental evidence for the study of hyperuricemia.Methods:Zhijuzi was extracted with water,50%ethanol,70%ethanol,and 95%ethanol respectively.The content of flavonoid constituents in each extract was measured,and the effect and kinetic model of the different solvent extracts on XOD were analyzed by ultraviolet spectrophotometry.Results:The extracts of Zhijuzi with different solvents all had inhibitory effects on XOD,among which the inhibitory effect of 50%and 70%ethanol extracts were relatively strong,and the contents of flavonoids in the extracts were also higher,and the inhibitory effect was dose-dependent.Although the total amount of flavonoids in 95%ethanol extract was lower,it also showed good inhibitory effect.By kinetic analysis,the affinity value α of different solvent extracts for XOD was greater than 1,and the affinity value of 70%ethanol extract for XOD was 134.40,which was more easier to bind to XOD.All extracts showed mixed inhibition.Conclusions:Zhijuzi extract could inhibit XOD,and the 50%and 70%ethanol extracts were more effective.The 95%ethanol extract was associated with other components in the extract.The type of inhibition of Zhijuzi extract was mixed inhibition,that was,competitive-noncompetitive inhibition,and the competitive inhibition was dominant.
