Effect of Electroacupuncture at Shenshu Point on Expression of M1 Receptor in Retina of Guinea Pigs with Kidney Yang Deficiency Myopia
Objective:To observe the effect of electroacupuncture at Shenshu point(BL23)on the growth,vision and expression of retinal muscarinic acetylcholine(mAChR)M1 receptor in guinea pigs with kidney yang deficiency myopia(KYDM),so as to provide a basis for the treatment of kidney yang deficiency myopia.Methods:Total 60 guinea pigs were randomly divided into normal control(NC)group,lens-induced myopia(LIM)group,KYDM+sham acupoint(KYDM+SHAM)group and KYDM+electroacupuncture(KYDM+EA)group,with 15 guinea pigs in each group.The NC group did not take any intervention measures.The LIM group was given a lens of-6.00 D in the right eye to construct the myopia model.The other two groups were given intraperitoneal injection of hydrocortisone(10 mg/kg)for 2 weeks on the basis of wearing glasses to construct the model of kidney yang deficiency myopia.After 2 weeks of modeling,the guinea pigs in the KYDM+EA group received electroacupuncture at bilateral Shenshu points,and the guinea pigs in the KYDM+SHAM group received electroacupuncture at the sham points of the buttocks for 4 weeks.During the modeling period,the general conditions of the guinea pigs in each group were observed,body weight and anal temperature were measured,binocular refraction and axial length were detected,serum free triiodothyronine(FT3),free thyroxine(FT4),testosterone(T)and estradiol(E2)were detected,and the mRNA and protein expression levels of M1 receptor in the retina were detected by quantitative polymerase chain reaction(Q-PCR),immunohistochemistry,and Western blot after the end of modeling.Results:After 2 weeks of modeling,the guinea pigs in the KYDM+SHAM group and the KYDM+EA group showed symptoms of kidney yang deficiency,such as hair loss,lack of luster and emaciation,arched back and squinting eyes,tiredness and laziness,intolerance of cold and cold limbs,and the serum FT3,FT4 and T levels were lower than those in the LIM group,and the E2 level was higher than that in the LIM group.The body weight,anal temperature,and right eye refraction were lower than those in the LIM group,and the axial length was longer than that in the LIM group,with statistically significant differences(P<0.05).After 4 weeks of electroacupuncture,the symptoms of kidney yang deficiency in the KYDM+EA group were effectively improved,and the serum hormone levels were normal.Compared with the KYDM+SHAM group,the body weight,anal temperature,and right eye refraction in the KYDM+EA group were significantly increased,the right eye axial length was shortened,and the expression levels of retinal M1 receptor mRNA and protein were decreased,with statistically significant differences(P<0.05).Compared with LIM group,the body weight of guinea pigs in KYDM+EA group was increased,with statistically significant differences(P<0.05);There was no significant difference in anal temperature,right eye refraction,axial length,and retinal M1 receptor mRNA and protein expression levels(P>0.05).Conclusions:Electroacupuncture at Shenshu point is beneficial to the growth,development recovery and vision improvement of guinea pigs with kidney yang deficiency myopia,and the mechanism may be related to the reduction of M1 receptor expression in the retina.
