Mechanism Study on Effect of Opposing Needling at Yanglingquan Point on Motor Function in Model Rats with Middle Cerebral Artery Occlusion
Objective:To observe the therapeutic effect of opposing needling at Yanglingquan acupoint(GB34)on motor dysfunction and the effect on callus myelin basic protein(MBP)in model rats of middle cerebral artery occlusion(MCAO),and to explore the mechanism of opposing needling at Yanglingquan acupoint to improve motor dysfunction of cerebral infarction.Methods:Total 68 male SD rats were randomly divided into sham operation group,model group,opposing needling group and non-acupoint group,with 17 rats in each group.Except for the sham operation group,rats in the other three groups were given thread embolization to prepare model rats with right MCAO.The intervention began on the second day after the operation.The opposing needling group was given electroacupuncture stimulation at right Yanglingquan acupoint,and the non-acupoint group was given electroacupuncture stimulation at bilateral non-acupoint under the lower abdomen,once a day,20 min each time,for 14 consecutive days.The grip strength and neurological deficit scores of rats were measured.The percentage of cerebral infarction volume was calculated by using 2,3,5-triphenyltetrazolium chloride(TTC)staining,the morphology of corpus callosum was observed by using HE staining,and the expression levels of MBP protein and mRNA in the right corpus callosum were detected by immunohistochemistry,enzyme-linked immunosorbent assay(ELISA),Western blot(WB)and reverse transcription polymerase chain reaction(RT-PCR).Results:Compared with the sham operation group,the grip strength was decreased,the neurological deficit scores were increased,the percentage of cerebral infarction volume was increased,the structure of the corpus callosum myelin was disintegrated,and the expression levels of MBP protein and mRNA in the right corpus callosum were decreased in the model group,with statistically significant differences(P<0.05).Compared with the model group and non-acupoint group,the grip strength was increased,the neurological deficit scores were decreased,the percentage of cerebral infarction volume was decreased,the structure of the corpus callosum myelin was intact,and the expression levels of MBP protein and mRNA in the right corpus callosum were increased in the opposing needling group,with statistically significant differences(P<0.05).Conclusion:The opposing needling at Yanglingquan point may promote the motor function rehabilitation of rats with MCAO by increasing the expression level of MBP protein in the corpus callosum.
