Analysis of Effect and Mechanism of Acupuncture and Moxibustion Preconditioning on Rats with Acute Gastric Mucosal Injury Based on Nrf2/HO-1 Pathway
Objective:To observe the effect of acu-puncture and moxibustion preconditioning on rats with acute gastric mucosal injury,and explore its mechanism.Methods:A total of 52 SPF rats were randomly divided into normal group,model group,moxibustion preconditioning group and acupuncture precon-ditioning group,with 13 rats in each group.Rats in the moxibustion preconditioning group and acupuncture preconditioning group were preconditioned with moxibustion and acupuncture at Zhongwan(RN12)and Zusanli(ST36),20 minutes each time,once a day,for 7 consecutive days.Then,rats in the model group,the moxi-bustion preconditioning group and acupuncture preconditioning group were given anhydrous alcohol with aspirin suspension to prepare the model rats with acute gastric mucosal injury.Ulcer index(UI)and pathological changes were compared in each group.Enzyme linked immunosorbent assay(ELISA)was used to detect the content of malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GPX),and the levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in gastric mucosa of each group.The expressions of nuclear factor erythroid 2 related factor 2(Nrf2)and heme oxygenase-1(HO-1)protein were detected by immunohistochemistry.Results:Compared with the normal group,the UI in the model group was increased(P<0.01),and pathological changes such as congestion and erosion were found in the gastric mucosa of rats in the model group,suggesting that the model was successfully established.The content of serum SOD and GPX decreased(P<0.01),while the levels of serum MDA,TNF-α and IL-6,and protein expression of Nrf2 and HO-1 in gastric mucosa increased(P<0.01).Compared with the model group,the UI in the moxibustion preconditioning group and acupuncture preconditioning group were significantly lower(P<0.01).In the moxibustion preconditioning group,no obvious pathological changes were found in the gastric mucosa,while in the acupuncture preconditioning group,the gastric mucosa was mostly intact with only a small amount of red blood cell exudate.Compared with the model group,the serum SOD and GPX content in the moxibustion preconditioning group and acupuncture preconditioning group increased(P<0.01),while the levels of serum MDA,TNF-α and IL-6,and protein expression of Nrf2 and HO-1 in gastric mucosa decreased(P<0.05 or P<0.01).Compared with the acupuncture preconditioning group,the serum SOD and GPX content in the moxibustion preconditioning group increased(P<0.01),while the levels of serum MDA,TNF-α and IL-6,and protein expression of Nrf2 and HO-1 in gastric mucosa decreased(P<0.05 or P<0.01).Conclusions:Acupuncture and moxibustion preconditioning can prevent the pathological damage of acute gastric mucosa in rats,and its mechanism may be related to regulating the Nrf2/HO-l pathway,alleviating oxidative stress and reducing inflammatory response.
