HPLC转换波长法测定木瓜中绿原酸、齐墩果酸和熊果酸含量

目的:建立HPLC转换波长法,同时测定木瓜中绿原酸、齐墩果酸、熊果酸含量.方法:采用HPLC法,Kromasoil C18(250 mm×4.6 mm,5 μm)色谱柱,流动相:乙腈-0.6%磷酸溶液,梯度洗脱,流速:0.8 mL/min,检测波长:326 nm,210nm;柱温:30℃.结果:绿原酸在0.192~2.880 μg(r=0.9994),齐墩果酸在0.488~7.320 μg(r=0.9999),熊果酸在0.437~6.468 μg(r=1)范围内,线性关系良好;三种化合物的平均回收率为别为101.13%(RSD=1.98%,n=6)、99.64% (RSD=1.97%,n=6)及99.66%(RSD=1.90%,n=6).结论:该方法简便准确,重现性好,可为控制木瓜质量提供参考.
Determination of Chlorogenic Acid、Oleanolic Acid and Ursolic Acid in Fructus Chaenomelis by Wavelength Conversion-HPLC
Objective:To establish a wavelength conversion method of HPLC for simultaneous determination of chlorogenic acid,oleanolic acid and ursolic acid in Fructus Chaenomelis.Methods:Stationary phase was Kro masoil C18(250mm×4.6 mm,5 μm) with the column temperature of 30 ℃.Acetonitril-0.6% H3PO4 was used as the mobile phase with gradient elution.The flow rate was 0.8 mL/min.The detection wavelength was 326 nm and 210 nm.Results:The chlorogenic acid had a good linearity relationship(r=0.9994,n=6) between 0.192~2.880 μg.The oleanolic acid had a linear range between 0.4887.320 μg with r=O.9999(n=6).The ursolic acid had a linear range between 0.437~6.468 μμg with r=1(n=6).The average recovery of chlorogenic acid was 101.13% with RSD of 1.98%,the oleanolic acid was 99.64% with RSD of 1.97%,the ursolic acid was 99.66% with RSD of 1.90%.Conclusion:This method is simple,accurate,reproducible and can be used for the quality control of Fructus Chaenomelis.
