紫叶矮樱离体快繁技术研究

Studies on Rapid Propagation in vitro of Prunus × Cistena

以紫叶矮樱的带芽茎段为外植体进行快繁技术研究,结果表明,外植体消毒采用5% NaClO处理20 min的效果最好,3月下旬到4月中旬为最适取材季节,启动培养以1/2 MS+ 6-BA 1.5 mg/L+ NAA 0.05 mg/L为最佳培养基配方;生根培养最佳培养基组合为1/2 MS+ IBA 1.0 mg/L+ IAA 1.0 mg/L,其次为1/2 MS+ NAA1.5 mg/L.1周时,最佳生根培养基组合1/2 MS+ IBA 1.0 mg/L+ IAA 1.0 mg/L有根长出,20 d后平均生根数达2.0~5.4条,生根率为48.6%~94.7%.
Prunus x Cistena with apical bud stem segment as explants was chosen in the plant nursery. Results showed that, explants were sterilized with 5% NaCIO adding a droplet TWEEN80 for 20 minutes, which were regarded as the best effectiveness. It was the most suitable for material selection from the middle ten days of March to the middle ten days of April. For the initiation culture, 1/2 MS + 6-BA 1.5 mg/L + NAA 0.05 mg/L was selected as the best combination incubation media recipe. For the rooting culture, the best incubation media was 1/2 MS + IBA 1.0 mg/L + IAA 1.0 mg/L, the better was 1/2 MS + NAA 1.5 mg/L. Root outgrew for 1/2 MS + IBA 1.0 mg/L + IAA 1.0 mg/L in one week, the average root numbers were in the range of 2.0 ~ 5.4 in 20 days, the rate of root was 48.6% to 94.7%.
