对根癌农杆菌介导的马铃薯遗传转化过程中涉及的外植体选择、预培养、共培养时间等因素进行优化.结果表明,马铃薯的茎段和叶片均能诱导出愈伤组织并分化成苗;在转化前对外植体预培养2d,再用OD600为0.5的农杆菌菌液侵染10 min,共培养3d,转化效率较高.利用优化的农杆菌介导法进行转化,对再生苗进行PCR检测,结果表明,特异切割马铃薯纺锤块茎类病毒的核酶基因已导入了马铃薯.
Influencing Factors to Potato Transformation Mediated by Agrobacterium
In this paper,the effects of different explants,the durations of pre-culture and co-culture on transformation frequency mediated by Agrobacterium were studied.The results showed that calli could be induced from both leaves and stems of potatoes and that optimized system was pre-cultured of explants for 2 days before transformation,infecting explants by Agrobacterium with 0.5OD600 for 10 min,co-culture of explants and Agrobacterium for 3 days.A ribozyme gene targeting against two sites on negative strand of potato spindle tuber viroid (PSTVd) was transformed into potato by this optimized system.
